木豆叶芪类提取物对雌激素缺乏性大鼠骨质丢失的影响

Abstract: The Cajanus cajan L. is a natural p lant, which contains lots of potential activecomponents. The effects of the stilbene extracts from Cajanus cajan L. ( sECC) on ovariectomy (OVX) 2induced bone loss in ratswere identified. All experimental female rats were divided into 6 group s, i. e. ,sham2operated rats, OVX rats, 17β2estradiol ( E2 ) 2treated rats, sECC2treated ratswith three dosages, 50,100, and 200 mg·kg- 1 , separately. Two weeks after the operation, different dosage of sECC, E2 ordeionized water were given to the 6 group s of rats, respectively for another 8 weeks through stomach.Then, all ratswere killed. The bodyweight and uteruswetweightwere measured. Contents of serum E2 ,follicle stimulating hormone ( FSH) , and luteinizing hormone (LH) were measured by radioimmunoassay.Femoralmorphology was observed by HE stain. The results showed that there were no changes of theuterine weight and serum E2 concentration in sECC2treated rats compared with OVX rats. However, theserum FSH and LH concentrations reduced by 1115% and 1512% ( P < 0105 ) , respectively. By HEstaining, it is found that the 60% of the femur structure had been significantly imp roved in OVX ratstreated with 200 mg·kg- 1 of sECC. The trabeculae were thicker and larger than that of OVX rats. It is clear that sECC imp roved femoral morphological structure and decreased FSH and LH contents without affecting serum E2 level and uterine weight in OVX rats. The results suggested that sECC had potential action in treatment of postmenopausal osteoporosis.
讨论 本研究中, 17β2雌二醇组的剂量是常规给药的一半[ 7 ] ,但仍能明显增加血清17β2雌二醇水平,子宫重量也明显增加。然而,给予木豆叶芪类提取物后,血清中17β2雌二醇水平没有明显变化,但可降低FSH及LH水平。结果表明,木豆叶芪类提取物具有类似17β2雌二醇的作用,逆转了因17β2雌二醇缺乏所引起的FSH及LH的升高,且不增加子宫重量,这一表现与补充雌二醇所产生的作用明显不同[ 8 ] ,但与已知的植物雌激素作用类似[ 9, 10 ] 。更有意义的是,中、高剂量的木豆叶芪类提取物可以剂量依赖性地抑制大鼠股骨头部位的骨丢失。表现为明显增加大鼠骨小梁数量、改善骨小梁的结构,与雌激素的作用接近。根据本研究中木豆叶芪类提取物所表现出的类似植物雌激素样的作用可以推测,在用于雌激素缺
乏所引起的骨丢失时,木豆叶芪类提取物能够在改善骨小梁结构的同时,不发生雌激素替代治疗时可
能出现的子宫癌和乳腺癌的危险[ 8 ] 。结论因此,木豆叶芪类提取物具有良好的药用开发前景。
